I've never used the variable wizard, so I can't speak to that - but if you have two processes - one with and one without the prime pillar - go to the ADVANCED tab to set the start and stop heights for each process.

Using a .4mm nozzle, I'd go to a .5mm extrusion width. The plastic expands a little coming out of the nozzle, so if you are running an extrusion width equal to your nozzle diameter, the plastic stream needs to be stretched a little during printing. 20% over the nozzle diameter seems to be a good num...
