I want to use simplify3d printing to Makerbot Replicator 5th Generation.

Should I move the .gcode file, every time by using USB, from my computer to the printer?

Are there any other convenient, direct way?

You can print over USB or just copy the .makerbot file to the SD card. Either method works.
