Custom Wall Thickness By Feature

It would be very valuable to be able to set wall thicknesses for specific model features. For example, I have a part that has holes for self tapping screws. It would be nice to thicken just the inside walls of these holes to accommodate the screw threads while leaving the rest of the model walls the global set thickness. Implementing this feature would have to identify features from mesh triangles or maybe not - lasso triangles directly? - anyway, that's where coding creativity and utility come in.

What you are talking is "different shells for different parts of the print", at the moment you can do this with Simplify3D using multiple processes. Create a separate process that has more shells and assign it to part where you want "thickness increased". You will also have to "split" your part in CAD software so instead of one part you import multipart into Simplify3D so you can assign different processes to different parts. You can cut your part in meshmixer if you only have STL or in whatever your source CAD is if you are designing the part yourself.

Customizable or variable wall thinkness is something I'm looking at as well
modern high-flow printers allow obtainig, with a fixed nozle, different thikness of "wall" just changing extrusion parameters, this in within a range of many mm.
This grants quick and extremely solid builds, but I'm able to play only along Z axis.
Expecially on shell "Vase" constructs, being able to change extrusion parameters on a single plane would allow extremely smoot transitions of thickness while conserving high strength and speed of build; palying with infill or multiple boundaries would not grant the same outcome, expecially in terms of mechanical stability and resistance
